I find that I need to give myself a week or two's worth of riding in between adjustments (so, 60-100 miles for me, since a normal week is 30-50 miles). It is *very* easy to overadjust, so I have a rule of only one adjustment at a time. I also find that it's a good idea to take frequent breaks on a new bike. Nothing major, even 30s at a stoplight can be a long enough break. If there are no stop signs or stop lights to force quickie breaks, I might take a minute or so off the saddle about every 15 minutes.

If I fiddle more with my bike, or don't take enough breaks, I get uncomfortable very quickly, even if I'm in a good riding position. It takes time for my body to get used to a new bike or a new riding position, and at first the change can feel worse. More riding time means that shorter and less frequent breaks work better.

The more riding time you get on various bikes, the easier it is to feel the changes you need to make for everything to be right.
