Not a 34t with the Tiagra. The 32t is as far as the derailleur will handle. As it is, even with the B-screw adjusted so the top pulley doesn't tag the largest cog, if you back pedal in that gear there's a little bit of a "bumpy" feeling where the pulley and the cog are a little too close for comfort.
