I actually haven't laced the hub up to a 406 rim yet. They are being shipped to me as we speak. The hub is currently laced to a 26" (559) rim and I have not used it in it's current stale.
As for chainline, I cannot answer you specifically, but I can tell you that internal gear hub (IGH) and coaster brake cogs, if they are bigger than 16 teeth, are dished, so you can flip them over (assuming you can get that circlip off....) to adjust chainline. Another way to hack the chainline is the use of chainring spacers.
I have done this with my fixed gear and it works great.

Sheldon Brown did it to both his Nexus 7
and Nexus 8
so I'm guessing it should work?
