Setting up new Cassette for Steep Climbing

I'm interested in setting up an extra rear wheel with a 12-30, or a 12-32 cassette for steep mountain climbing. I currently run a Ultegra 11-27 10-spd, with a 53-39 double chainring, and a Ultegra RD for everyday use.

Can I easily interchange the extra 12-30 or 32 rear wheel without having to swap out my chain and or/ RD? Are there any pitfalls I may be getting into with this idea?
