I just picked up a cannondale, comfort 200. I'm wondering if a wheel set by Rolf. Vector pro we'll work now with this frame. Thank you for any help.

The Rolf rear wheel will be spaced at 130mm. I don’t know the cannondale’s specs off the top of my head, but being a comfort bike it likely takes a 135mm wheel.

Without more specific details on the bike I can't say much. But "Comfort 200" sure sounds like a hybrid/casual/cruising type bike and the Rolf Vector wheels are, IIRC, pretty racing like wheels and not intended for the type of use and tires the usual hybrid has. Beyond the rim width and tire width aspects I also suspect the rear wheel's axle OLD will be other than what the Cannondale frame is made for. If I'm right this sounds like wanting to run 40 series tires on your KIA mini van. Sure it can be done but why Andy

no... WAY too narrow, and the wrong rear axle length, AKA: Over Locknut Distance ( O.L.D. abbreviation )

here's the closest specs i could quickly dig up on the web... your bike even has a different wheel diameter than the Rolfs.
it has 26" MTB Wheels.. the Rolfs are 700c.
