Bike fit Question

I have a Specialized Roubaix Elite and in the present set up I'am comfortable anfd have no problems there but am would like any imput anyone may have on set-up. What I'am looking to achieve is maxium climbing ability within a comfortable set-up. Where should my seat alignment be ? (forward or back ) . My knee alignment to pedals ?
