The easy route is to get a set of cheap Open Pros, then pay ~$15/wheel with a good wheel builder to tension them properly, bonus if you get him or her to loosen the spokes enough to adjust the spoke line coming out of the flanges.
The suggestion of Velocity Fusions is a good one too. I built up a set of 32h/36h Deep-Vs, and the damned things will never die. Fusions are a little more sane, but still stonger than Open Pros. If you went with Fusions, I'd recommend 28h/32h. Aero spokes are fine, but don't make much difference.
Do check the bearing adjustment on Ultegra hubs. All the ones I've seen come over-tightened, but I've heard (as Brian_Ratliff mentioned) of some coming loose. Whatever the case, they're absolutely not going to be adjusted properly (so there's a touch of play with the QR open and none when it's closed).

You can get OP rims + Ultegra hubs complete wheels built for $200?? That's a killer price. That's like $50 cheaper than the cheapest on eBay !

Making a kilometer blurry
I don't mean to threadjack but I just got some new Ultegra 6600 hubs for a build (Aerohead O/C + DT butted). When I spin the rear hub (not the cassette body), I heard a screeching sound, almost like the rubber seal against the hub body. Does the hub need to be broken in, or is this a cone adjustment problem? Thanks for the heads up on the bearing adjustment... I would have assumed it was fine.

Squeaking of the freehub seal will probably go away. To see if your hub is adjusted right, open the QR so that it's just strong enough to hold the wheel in (maybe 45 degrees open). Wiggle the wheel from the rim. You should feel some play. Re-tighten the QR and the play should be gone. I often fiddle slightly with the QR tension to refine the play, but you don't want to make up too much ground there (for safety).
