Cassette spacer -when needed?

How do you tell when you need a spacer on the cassette? For my 9 speed touring setup, I am putting a Shimano 11-34T cassette on a 105 hub (5600). A quick fit check does not show any problems but I know that a spacer behind the cassette is needed in some cases. As long as I can dial in my rear derailure, does that tell me that no spacer is needed?
