Personally, I prefer to wear something that wicks away any moisture and dry quickly. Modal, microfiber (under armour), coolmax (TravelSmith, Calvin Klien, Nautica, etc), Supplex, Polypro, etc etc all do the job and help prevent chafing and blisters/sores. Silk underwear is harder to find, but some prefer natural materials and feel they are more comfortable yet wick away moisture and dry quickly. A lot of swim trunks and board shorts have coolmax linings/support and that can be a viable alternative.
Udder Balm (cream) and Gold Bond (powder) have been used for years and still work. Runner's Lube/Lube-Stick or Body Glide are also popular.

I drive a semi all night and cotton sport briefs are absolutely hellish. I've used these for short rides to 30 miles or so with my Brooks B67's. Ordinary nylon baggies outer-side. Seem to be ok, but nothing beats a pair of padded, non-gel spandex bike shorts, commando.

This "underwear - no underwear" discussion can go on forever, however, it's one of those decisions we all must make on our own. Many people use these cycling undies that have some padding, and add just a bit of comfort. See for yourself, they are actually very comfortable.
