Why not? Just be sure to protect the disc on the removed wheel from getting bent. They're not like delicate butterflies, but if not handled with care the disc can get bent and you won't know it until you remount the wheel and hear it rub-rub-rub-rub. Not that I've ever done that.

Yes, but if the brakes are hydraulic you have to be careful not to operate the brake lever with no disc rotor in place. You can prevent any problems by inserting a spacer into the place where the rotor fits through the calliper, these are used on bikes being shipped to bike shops in their boxes. When I worked in a shop, I had a drawer full of them, I only have 2 left. I use one each time I remove a wheel to transport my bike in my car

I use a composite door shim (from the hardware store) in my caliper when the wheel is off...Use a piece of blue masking tape to keep it from falling out. My LBS didn't have the aforementioned shipping spacer.

you put it in the caliper when you take the wheel out.
but you just didn't know to ask Them, for it, when you bought the bike..
now go back and get it.

its not rocket science-I usually fold a piece of thick paper, or thin cardboard(matchbook cover), and stick between brake pads-you can take front or rear wheel off just as you do with any other bike.
most bike shops will have several spacers laying around and will hand them out if asked, I have a couple on my bike stand I use when washing bike

When you take off the front wheel, make sure you store it in a safe place that won't bang into anything while you're in the car. Even an old bath towel can be used to cover it during transporting.
